SHLP6 (Small Humanin-Like Peptide 6) is a 20-amino acid mitochondria-encoded peptide from the 12S rRNA gene region, identified alongside SHLP2 and the other SHLP family members in the 2016 USC discovery. SHLP6 is notable for a unique dual biological profile that distinguishes it from all other mitochondrial-derived peptides: it is cytoprotective in normal, healthy cells — protecting them from oxidative damage and stress-induced death — while demonstrating pro-apoptotic activity in cancer cell lines, selectively triggering programmed death in transformed cells. This cancer-selective pro-apoptotic property is not shared by Humanin, MOTS-c, SS-31, or SHLP2, and makes SHLP6 the subject of distinct oncological and cytoprotection research tracks.
