SS-31 (elamipretide) repairs the inner mitochondrial membrane by binding and stabilizing cardiolipin — the phospholipid that holds the electron transport chain in place. When cardiolipin degrades with age, electron flow becomes inefficient and reactive oxygen species production spikes. SS-31 fixes this structural problem first. MOTS-c then acts as the metabolic reprogramming signal — a mitochondria-encoded peptide that activates AMPK, driving mitochondrial biogenesis, glucose uptake, and fat oxidation. Finally, NAD+ supplies the essential coenzyme substrate that every mitochondrial energy process depends on, restoring the declining NAD+ pool that drops approximately 50% between ages 40 and 60.
