BT-11 is a gut-restricted, orally bioavailable small molecule peptide ligand developed by Landos Biopharma that selectively activates the LANCL2 (LanC Like Protein 2) signaling pathway in intestinal immune cells. BT-11 is specifically engineered for gut lumen restriction — it does not cross the intestinal epithelium and achieves no meaningful systemic absorption, making it a uniquely targeted gut immune modulator that avoids systemic side effects. LANCL2 is a mammalian homologue of bacterial LanC enzymes that functions as a receptor for abscisic acid (ABA), a plant hormone also produced in human immune cells. LANCL2 activation drives immunoregulatory effects including increased IL-10 production, reduced IL-17 and TNF-alpha secretion, and promotion of T-regulatory cell function in the gut-associated lymphoid tissue (GALT) — recalibrating the hyperactive mucosal immune responses driving IBD and Crohn's disease. Phase 2 clinical trials for Crohn's disease demonstrated favorable safety, reduced fecal calprotectin (an inflammation marker), and symptom improvement.
