ARA-290 binding to the IRR activates anti-inflammatory pathways including inhibition of NF-kB, reduction of pro-inflammatory cytokines (IL-1beta, TNF-alpha, IL-6), and stimulation of tissue repair signaling without thrombotic or erythropoietic side effects. FDA Fast-Track designation was granted for autoimmune small fiber neuropathy (SFN) — a painful peripheral nerve condition — following Phase 2b clinical trials demonstrating improved corneal nerve fiber density (a proxy for SFN severity) and symptom reduction.
