LEAP2: 5mg
Weight managementDescription
Leap2 (liver-expressed antimicrobial peptide 2) is an endogenous circulating peptide produced primarily by the liver and small intestine that functions as a competitive antagonist and inverse agonist at the growth hormone secretagogue receptor (ghsr), the receptor for the orexigenic hormone ghrelin. by opposing ghrelin signaling in hypothalamic and brainstem circuits that regulate appetite and reward, leap2 has emerged as a mechanistic target for obesity and metabolic research distinct from glp-1, gip, glucagon, and amylin pathways. in lean healthy men, earlier infusion work showed reduced food intake and glucose-lowering effects; englund et al. extended this to men with obesity in a randomized, double-blind, placebo-controlled crossover study published in diabetes (2026;75(6):925–937; pmid 41911360). continuous intravenous leap2 at approximately 40 pmol/kg/min raised plasma leap2 about fivefold versus placebo, reduced ad libitum energy intake by roughly 12% (mean −411 kj), and attenuated postprandial glucose peak and auc during a mixed-meal test. ongoing and related human protocols include leap2fmri (nct06845033) examining food-cue reactivity with fmri and silenced (nct07171723) testing leap2 during stable semaglutide treatment to probe ghrelin-axis interactions under glp-1 therapy. leap2 remains investigational with no fda-approved product and no established subcutaneous research-vial dosing paradigm in the peer-reviewed literature. for research and educational purposes only. not fda-approved. consult a qualified healthcare professional.
Regulatory Notice (October 2026): LEAP2 (liver-expressed antimicrobial peptide 2) is an endogenous ghrelin-receptor antagonist/inverse agonist under academic clinical investigation. It is not FDA approved, is not a component of any FDA-approved drug product, and does not appear on the 503A or 503B bulk drug substances lists. Human studies to date (e.g. Englund et al., Diabetes 2026; ClinicalTrials.gov NCT06845033, NCT07171723) have used intravenous infusion protocols for mechanistic research, not pharmacy compounding. Research compound only. Sources: Diabetes 2026;75(6):925–937 (doi:10.2337/db25-1132; PMID 41911360); ClinicalTrials.gov NCT06845033; ClinicalTrials.gov NCT07171723.
